Laboratory 6 and 7:  Patient Unknowns for Gram positives and fastidious Gram negatives, Throat Cultures

Activities:

  • Throat Cultures
  • Patient Culture Unknowns
  • Plates of Known Organisms :Review both the gram positive and gram negative fastidious organisms and fill out the appropriate worksheets
  • Set-up Demo Tests
  • Read Culture Unknowns (Unknown #1 and throat culture)
  • Set up Identification Tests and Susceptibilities if appropriate
  • Read Demo Tests with instructor

Throat Culture

  1. Collect throat cultures with your partner.

  2. Streak BAP, SXT, and Chocolate plates on Wednesday following the classic 4 quadrant streak pattern. 

  3. On Thursday, describe colonial morphology,  Gram stain, and perform rapid tests on the different colony types using the throat culture worksheet.

  4. Complete the Throat Culture Worksheet

Patient Culture Unknowns

  1. Plate your  unknown on the appropriate media using your lab manual as a guide.

  2. Work up your unknown using your flow diagram for gram positive organisms on Thursday and Friday.   Check with your instructor on whether you will be setting up a gram positive susceptibility on your unknown.

  3. Complete the Spinal Fluid Culture Study Guide

Plates of Known Organisms

Blood and Chocolate Agar Plates are available:

  • Neisseria gonorrhoeae

  • Neisseria meningitidis

  • Neisseria lactamica

  • Neisseria sicca

  • Moraxella catarrhalis

  • Haemophilus influenzae

  • Haemophilus parahaemolyticus

  • Haemophilus parainfluenzae
