UNIT I PRACTICAL REVIEW
- Perform a Gram Stain on a prepared slide.
- Streak plate for isolation and preparing a Gram stain(s) from the
culture (refer to last weeks evaluation of streaking for the grading
process)
- You will be streaking a plate from a prepared swab on Thursday. During
the lab practical you will evaluate the plate you streaked the day before
and make Gram stain(s) on the colonies on the plate. I will read the Gram
stains you have prepared and stained.
- Recognize type of hemolysis and swarming.
- What grows on each of the following media and differential reactions if any:
- Columbia CNA agar
- SXT agar
- Bile Esculin Agar
- Chocolate agar only
- Modified Thayer Martin agar
- MacConkey
- Eosin Methylene Blue agar
- Tergitol 7 agar with TTC
- On Tergitol how does E.coli look different than the other lactose
fermenters?
- Distinguish between the following growth requirements:
- Obligate aerobe
- Obligate anaerobe
- Capnophilic
- Perform a wet mount and distinguish between cocci and yeast at 40X
- Perform the following rapid tests from memory:
- Catalase
- Oxidase
- Coagulase
- Spot indole
- When given a Gram stain to read, determine when to perform an oxidase or
catalase and then perform it if asked.
- Read a bile solubility
- Know the reagents needed for the following:
- Catalase
- Oxidase
- Coagulase
- Spot indole
- Bile solubility
- Read provided Gram stains for:
- Gram stain morphology
- Extra credit for indicating what the organism might be:
- Quality of sputum under low power evaluation using Bartlett’s
classification
